Only Agents can configure the help desk. Access the project through JIRA's interface, go into Permissions and roles for the project. Make sure you are in the "Administrators" role of the project. You can also double check if you have the "Manage Agents" option when you click on "Service Desk" in the top panel (that means you have the permission to add yourself as an Agent to the SDs)
