is it possible to connect single database to multiple jira instances ?

I tried to Connect two jira instances to single database i got an error like  "You have specified a database that is not empty, please specify an empty database".

It sounds like you have not set up the Jira systems to be nodes of a Jira cluster properly.  You should have another read of the documentation about adding nodes to a Jira Data Centre installation.
