How to make custom fields as uneditable once the value is entered. Thus allowing only one time value entry.

I'd consider how annoying a user would find this. Seems arbitrary.

If you only want to allow it to be entered when an issue is created do as Jo-Anne says.

The easiest way would be to simply not put them on your edit screen.

You need to create a new custom field type for this. Or maybe handle it in Javascript.
