We would like to use a custom "People" field to essentially add multiple assignees to a particular issue and using automation rules, email them regarding any updates to the issue.
How would it be possible?

I don't believe those types of custom fields are available to Automation For Jira for Team-managed projects yet. You might want to click on the Give Feedback link on the bottom left of the project menu and let them know you would like to have it.
Yes, and...to John's answer: Here are a couple of the open suggestions to make People fields available in automation rules.
