Can anyone tell me how (if possible) to desginate a custom field as a mandatory field for an Issue in a Next Gen project?

... and no; we have NO intention of using Classic projects. :-)

No, not yet.  There is a suggestion to add that feature, which you may view and vote for here:

Only a few built-in fields support required at this time, using a validator on a project's workflow.
