Can you guide on how to set up for developers who are sometimes requested to work over the weekend. How does one define a Compensatory off for the same work on the weekend on another day?

In terms of HR? You need to make sure that this is clearly defined in the employees' contracts.
In terms of Jira Cloud, I would use the Tempo plug-ins for planning and time entry (https://marketplace.atlassian.com/vendors/6558/tempo-for-jira)
Under your Overheads project (or whatever that is called) create an issue titled "Time Off In Lieu".
That way you can plan, record, and monitor the compensatory time off employees have earned through weekend work. You can also link these issues directly to the worked issues so it is clear what they did and for how long.
