Is there a way to create epics in Jira from email who are not registered in JIRA?
For example, I want to streamline request from external clients. Clients would send an email with required information to the associated jira service email. Jira would then create an epic and place it in the backlog.

You may do that by setting up a mail box user and Jira account just for receiving new issues. Please look here for details on that approach:

Thank you so much! Another question I have is if there is a way to auto populate special fields in the issue. Like a pre-made template that would populate information into certain fields.
Example:
Request Title -> Summary
Request -> Description
Country/Region ->Special Field
Business Unit -> Special Field

Yes, that completely makes sense, and I do not believe that is an out-of-the-box feature. There are most likely marketplace add-ons for purchase to do this, with template creation features.
As a work-around, you may use Automation for Jira (A4J) to "catch" when an issue is created and then add the new field values you wish. If you are interested in trying that, here is some documentation and examples of what is possible:
