I have gone through existing discussions as how to update existing records in JIRA by using Import option with admin rights. My question is, instead of using 'Issue Key' field in JIRA, can we tell JIRA to use field from our import file , as Key field to search and update JIRA record.

No.  Jira relies on there being a guaranteed unique way to identify the Jira issue you want to update.  The only uniqueness that Jira can always rely on is its own, so the issue Key (or id in the database).

It can never be sure that any reference you give it, other than the key, is unique.
