Is it possible to set the assignee field mandatory in Jira?

No, you can't set it as mandatory. You can set "allow unassigned issues" off, which makes it globally mandatory. And you can use validators to force it to be entered before proceeding through the workflow. But you can't quite make it mandatory in the standard way.
