I'm curious to know that how the tagging is working in jira comments, how they are hanlde mentions

I want to know that how tagged one can differentiate the mentioned users from each other

Welcome to the Atlassian Community!

I do not quite understand the question.  If you tag someone in a comment in a Jira issue, then they are tagged, and will be told they have been mentioned (via email). 

I'm not sure what you are looking for when you say "differentiate the mentioned users from each other".
