Each and everytime I open JIRA, its shows JIRA Startup Failed, JIRA has been locked.

This think is being happened after I developed and install a plugin.

Tthe message being show, when I open JIRA...

Look at the log file - that will tell you why it's not starting all the plugins.

Two main culprits to look for though - invalid version of Java (should be 1.6) or having anti-virus software actively scanning the Jira directories (I've only seen this be a problem on Windows servers).
