after installation you can go to Administration section -> System -> REST API Browser

But, my query is how to identify if plugin Rest API browser is being used or not in jira? And, how to check data associated with this plugin?

This plugin is just a way to interact with builtin apis provided by JIRA. REST APIs are automatically provided by JIRA and there are no other APIs provided by that plugin.
