Our current set up for our helpdesk on JIRA. Is if we comment on a job, it sends a email out to the customer from a Jira@ address. This is a NoReply Email.
Is there a way to set this up so that if someone does get a reply. They either receive a email back with a message saying that the email will not be received and to email a different address. Or to have their reply forwarded to a specific address?

The idea of having your “NoReply” email address response with a “no one is home” email is a great idea and can avoid a lot of confusion when people do reply to the emails sent out by Jira. The good news is there is a feature request already submitted for this item. You may find this feature request at JRACLOUD-45249. We would suggest you vote and watch this request to receive future updates on its status.
