How Kanban columns like Development and Testing can be broken down to Doing and Done stages, as an example, is this possible in JIRA?
Jira doesn’t have the concept of sub statuses. To do this you would just have to leverage statuses and names that makes sense to you. for example - dev-doing, dev-done.

It sounds like you want to create a Pull System for your Kanban board and process. What we have done is created two statuses in the workflow for each activity. One called Underway and one called Pullable. So in your case, one for Development Underway (or Develop Underway if you want to shorten it a bit) and one called Development Pullable.
Create a transition from Underway to Pullable statuses and label the transition Make Pullable.
Then in your board, put the Underway status in the Doing column and the Pullable status in the Done column. Repent for both Development and Testing.

I was wondering if you know if the the "pullable" term is widely used in agile circles. I ask because my team is adopting this workflow and I hadn't seen it before and found it a bit confusing. Unfortunately, I can't think of an obvious substitute that doesn't have its own problems. If "pullable" is common lingo, I would be curious to know.

Kanban is a Pull system. But just because a card is in a column doesn't mean it is ready to be pulled to the next column. Once the criteria is met, then it becomes Pullable, meaning it is ready to be pulled. You wouldn't say it's become Pull. Probably just a word semantics thing.
