Can I set a project key for a specific issue type in Jira-Service-Desk?

Is it possible to have separate issue key format for specific issue type? 

Following is the scenario - 

Project is Nanitwo Project and key is NTO

I have issues types : Defect, Enhancement and New-feature.

I want the issue key for Defect as NTO-DEF-1 where as the issue key for Enhancement as NTO-ENC-1  and for the New-Feature as NTO-NF-1

Is this possible using any custom code? If yes can you list few of the options if you have tried this anytime?

this is not possible the project key is for the whole project.

Like Jack said.  You could get a 'new' project key for a specific issue type by creating a new Jira project and having that issue type available there.
