Autoasign tickets to Active users when ticket responsible gets disabled

We need this to happen  could this be posible?

I don't know what you mean by "when ticket responsible gets disabled". Can you provide more details on what that means?

It is possible to use Automation Rules to automatically set the Assignee field for issues, but I'm not sure if the rules can accommodate the condition you mention until you provide more info about what that means.
