@Alex you have two options.  

You can turn off allow unassigned issues in the general configuration.  Which will make it required on all issues and projects in Jira.

That's another way to do it, yes, but it's a bit clunky.

Go to project settings and look for the "field configuration scheme".   

Go into that, and look for the "field configuration" for the issue type you want to make it mandatory for (there may well only be one field configuration listed, in the default spot)

Go into the field configuration, you'll find it lists all the fields in the system (not just the ones in this project/issuetype).  Go find "assignee" and click the "mandatory" option to the right.

Be aware that field configurations are shared objects - there may be other projects and issue types using this one.

This won't work per OPs screenshots the required link is missing for the assignee field.
