I do not want story point estimates to be a mandatory field for when tickets are being created.
I would however like to know if, once a ticket changes from open to groomed, if this field could be made mandatory?

You can do it using workflow validator.

Yes that can be done.
Check the field configuration of the issue type. If "story points" is required, make it optional.
Then go to the workflow transition, Open ---> Groomed . Add a transition screen which has the field "Story points". Add a workflow validator ( may be Field required validator ). You can make the field mandatory during the transition.
