1) How can I create custom required field only in story Issue and at the same time, the required field should not affect the subtask created in the story ?

2) Can I make Original Estimate field required in Jira Cloud ? I can't find it in Field Configurator

For #1 you will want to have different workflows and screen schemes for your story and your sub-task. This will allow you to configure them to behave differently and as desired. For #2, when do you want to make that field, required? Consider doing this using a validator on your workflow at the specific transition you desire.
