We would like to require a version number in order to move an issue/ticket into the "done" status column in Jira software. Is it possible to set this as a requirement in Jira?

Yes this can be done using the transition screen, just create a screen with the Fix version field on it and then add the screen to the transition going into the DONE status. This way you have a prompt asking for the version before moving into Done status.
Also you can use the validators in the workflow to make the field required and throw error if the field is not filled.
