This is probably a dumb question, but we'll see what I get for an answer as I'm curious how this works. Our Marketing division would like to have very customized settings put into Jira, so we were thinking they may be able to have a second instance of Jira PROD set up for them. This way we could isolate the customization settings they are requesting to their instance and it would cause residual outcomes in the other non-Marketing projects. Is this possible, or would this require us to buy an additional license?

Would additional licenses be required if multiple instances of the software were to be installed on a single server?
So yes, you would have to purchase an additional license if you want to run a second production environment (the part about the single server is not relevant here). I hope this answers your question.
