Welcome to the Atlassian community.
It is not possible to customize the subject for an internal (non-Customer) email notification template in Jira Cloud with native functionality. There is a change request about that here:

Your options are to use Automation Rules to send the email instead, or to use a third party app that provides more options for customizing emails.
