I have defined a card layout with the intension of allowing me to easy/quickly open sub-tasks and linked issues "directly from the card".
The layout is shown here:
The issues listed in "line B" are sub-tasks, and the issues listed in "line C" are linked issues..
However, I have been very dissapointed to observe, that the issue ID's listed in lines B and C are NOT active hyperlinks, but just pure text.
Consequently I cannot open the issues in question directly from the card.
Is there any way I can make them be hyperlinks?

Not as far as I know based on normal configuration.
Would the issue detail view not be sufficient for this though? This is the right-hand panel which appears when you select a card - in there should be your linked issues and sub-tasks.
If linked issues is missing at all (and you're not on Cloud's new view) - you can add it by:

Accessing sub-tasks and the linked issues from the Issue Detail panel is indeed a possible workaround.
I would, though, prefer to avoid "the step" of changing my focus to the Issue Detail pane, and scroll down to sub-tasks and links sections, which you most of the times needs to do (at least in the Jira setup I am working with).

I had a look through the options but it looks like items on the card are just text as standard - even URLs, as requested in JSWSERVER-16165
You could raise a suggestion for it if you think it would be something you and others would find useful? :)
