How do I ensure that a request made in the customer portal is automatically assigned to a specific user?
I want the user to be selected for their specific portal group.

The portal group is not stored anywhere and cannot be used to find out where the request were created, so your only option is to look at either the request type or the organization that the reporter belongs to in order to identify who should be assigned the request.

So can I create a custom field, where the user can choose an option and this will be the key to associating it with an agent? If yes, how can I do this?

Yes, that is possible. And then all you need is an automation that will set the assignee based on the value selected.
To create the field just go to Settings > Issues > Custom fields and create it from there.
