I have it setup such that e-mails to and from clients can be cc-ed to a JIRA e-mail and these get added to the issue comments if the issue key is stated in the e-mail subject line. I am trying to find a way to ensure if the issue key is not mentioned in the subject line, JIRA could use the sender's email and match it to the client's email field and find the correct issue that way.

That is not possible.
The processing of emails is a closed system in Jira. There is no option for you to change the configuration of the mail handlers in the manner you suggest.
