Can the Transition state be locked to a single workflow state?

I'm afraid that does not make sense in JIRA speak.  A status is a status, you use transitions to move issues from one status to another.  There's no "transition state", just transitions and status, and if you really mean "transition status", then that would translate to status anyway, so it would be the same thing.

Could you explain a little more?  Maybe with a worked example like "My issue is in status X, I use transition Y and it change to status Z - what I want to add to that (or change) is ..."
