Can I activate ranking while having a custom sort?

I currently have a custom sort on my board "ORDER by priority ASC, due ASC". However, I want to be able to quickly drag tasks from backlog into active sprints if something comes up that is urgent. How can I have the drag and drop (into current sprint) without losing custom sort?

No, your custom sort overrides the ranking.  You can't rank and simultaneously give the issues a different order with another field.

Thanks for the info. Is there an easy way to love items from the backlog to the sprint without having to change between ranking and the custom sort?

Not easily, as the sprints are entirely based on ranks.  You can drag things into a sprint or directly edit them and change the sprint one at a time.
