Is it possible to have an automation in which, when a specific comment/reply to a customer is added, the status will change from Waiting for Support to Resolved?

It probably is, depending on what you are trying to read out of the comments. Can you provide an exact example?
Sample:
If this comment were added it must update from Waiting for Support to Resolved.
Hello {{issue.reporter.firstname}},
You have been assigned a license. Please allow 10 minutes for this to take effect.
