You can also use custom fields and have a dropdown option for priorities if you wanted to use something besides labels, but generally, labels would be the right way to go.

@John McCabe labels are a good choice for prioritising because then you can filter the board to show labels of a given priority on both desktop and mobile

There is a power-up for this very function so you're in luck. From the Power-Ups menu you can find/add Card Priority by Screenful. Once that's enabled define the priorities, then you can open any card in the board and then set it by clicking on Card Priority.
