User means the employee who can login to Jira.
User tier means, maximum number of users who can login to Jira.

If you select Monthly billing it shows you how much the monthly price is per licensed user.
With this billing model, at the end of the billing cycle you get charged based on the number of licensed users you have at that moment. This allows for flexibility in your number of licensed users; you may have 100 licensed users at the end of one billing cycle and 75 at the end of the next billing cycle. In the first cycle you would be billed for 100 * 12.48. In the second billing cycle you would be billed for 75 * 12.48.
If you change it to Annual billing it tells you what the price is per year based on a User Tier.
For Annual billing with a User Tier you select the tier that includes the maximum number of licensed users you expect to have at any point during the billing period. That is usually based on how many licensed users you expect to support at the beginning of the contract and then a buffer to account for your user base growing. With a User Tier of 201-300 you could never have more than 300 licensed user for your system. You would be prevented from granting the 301st user access unless you removed access from another user. You will pay this amount for a year of using the product regardless of how you count of licensed users fluctuates over the year.
