Can I use a G Suite user alias for the custom email? I have an account, I am the G Suite admin, and I have an email alias for my account, support@mydomain.com. Can I login with that alias to Service Desk for the custom email? Thanks.

Reading the details you provided, I'm afraid to say that it's not recommended to use an alias as a custom email address for a Service Desk project.

On a project it's important to use an email that no one has access, it should be an email address exclusively for Jira, because in case someone reads the email before Jira, the ticket won't be created.

When connecting a custom email, Jira will read all emails in the inbox in order to create tickets or add comments to already created tickets, that's why it's important to add an exclusive email for that.
