Custom fields have been created in Jira but I'm not able to find them in the POST call API response.

The custom fields have been created but are not present in the API response call. How do we add the fields in the API response?

            "The field 'customfield_13406' is not valid for this request type 'General inquiries'."
and for another request type
            "The field 'customfield_13406' is not valid for this request type 'Service Request'."

Your errors are clear - those fields are not configured to be used on the request type you are trying to make.

You will need to ensure the new Custom Fields are accessible by the Projects and the Portal
