May I know is there a setting to disallow ticket to resolve if a field still not update?
As example, ticket cannot resolve if the value of Department field is Unknown. It can be resolve when change from Unknown to any other department.

Yes you need to put a condition on your transition.
Go to the Conditions section on the link below. You'll see an option for Value Field and then instructions on where to set the condition on your transition.
