I have set up Service Desk (only one) on my OnDemand JIRA instance. Customers can reach it by going to :
https://MyCompany.atlassian.net/servicedesk/customer/portal/2
Not really a user friendly url.
Is there a way to set up a redirect like https://MyCompany.atlassian.net/gethelp or even better https://gethelp.MyCompany.com (I control my DNS server).

So basically we need to do it ourselves outside of Jira, and that won't be persistent as soon as you follow a link inside the ticket.

If you control the domain, then you can put a simple redirect on there.
But it won't change the place Cloud services live at, so people will end up on the original url. Cloud does support changes of the <mycompany> part of the url, but you can't change any of the rest of it.
You could proxy and rewrite the whole lot on your web-server on the urls you want to land on, but Cloud is still going to refer to its naming scheme throughout the system.
