I am a VA for a small office that wants an easy way for employees to track PTO in one place.
I picked Trello because I thought they may want to use the boards for things other than PTO like workflow tracking.
How would I go about building a board for this office?

For tracking PTO, you might want to use the calendar power-up. If you're on the Premium plan with Trello, you have access to the built-in calendar view.
