I want to auto-assign the Jira ticket to someone else if the assignee is on leave. Is there any possibility of reading the leave date from the calendar and handling the same?

Unfortunately, it is not possible using the calendar managed in Confluence.
However, if your env is Data Center, then you may be want to take a look at using Automation for Jira to manage issue assignee using automation rules -

No need to access from the confluence. Currently we are adding our leave plan in confluence.

My company has an app for Jira Cloud that would suit your purpose. It's Out of Office Assistant for Jira Cloud.
We are building integrations so that vacation plans can automatically activate out of office rules, and then issues get reassigned to a different user who's backing the absentee.
Right now, you can integrate Outlook or Google Calendar through Zapier. I can give you a hand setting it up.
We're also working on an integration with Tempo so you can handle vacation plans over there.
If you have enough resources, you could even build your own integration with the OoO API.

We developed an app that solves this problem in a more comprehensive way, without changing the assignee. I suggest you take a look at Keychain - Delegation for Jira by OBSS.
With Keychain, you can set delegates for people and you can configure workflows and schemas that allow those delegates to perform actions.
