All:
The automatic backup (Confluence) assumes a lot of space and we are planning
to use server back up.
My question is:
1) Does the Database server backup really backup everything?
2) Should we back up anything else to make sure that the backup is complete.
3) Any restore process to check if the backup works perfectly.

Mostly, but it depends on your config
There's a small amount of configuration in the application directories you might want to think about keeping (database connection settings at least, but then if you've done any core hacks, changed properties files etc)
Are you keeping attachments in the database or on file system? If they're on disk, you'll need to back them up outside the database backup.
You might want to consider backing up the application home directory as well, as that can give you a quicker recovery during a disaster.
As for proving it - that's up to your backup routine. The only way you'll know if your backup works is to restore the whole lot to a temporary system and test it.
