I can't open .doc documents with the function "edit document" when Word 2016 ist installed on the client. The same document can be opened and changed with Word 2013 without problems. Do I have to change settings in word 2016?

Office 2016 is not yet officially supported in Edit in Office. Have a look here at this feature request:

I have heard a few users that have been able to get this to work - do you receive an error message when you attempt to open the document?

You can try the following, suggested from the improvement request:
In Word or Excel, whichever you're using, go to File>Options>Trust Center>Trust Center Settings>Protected View, and deselect the 3 boxes you find there.
You could also take the URL of the document from Confluence, and paste it in the Run menu on Windows. This may allow you to edit the file but you'll still need to save it locally and upload since indeed Office 2016 is not supported.

Confluence edit of Microsoft documents has been working for months until yesterday... Now, I am having the issue when trying to edit, the circle (image below) just continues to spin without end.

You'll want to check your Confluence server logs to see what's going on there. If you're using Confluence Cloud you'll need to raise a support ticket so we can look at your logs for you.
