I haven't worked with Scriptrunner to make a REST endpoint, but here are the docs in case you're searching for something specific.

In order to achive what you had described, I don't think you need to develop a REST Endpoint.
I guess the following procedure that doesn't require development:
Summary, Custom Field Nameopttion1, YOUROPTION1000FROMSAP...option1000, YOUROPTION1000FROMSAP
My idea is to simulate an import of tickets with new options of your custom field. If the option doesn't exist, it will be added in the import.

An app like Elements Connect might help you. Such apps enable you to show data from SQL Databases, REST APIs, or similar, as custom fields inside Jira.
I've used that app ages ago but back then it worked perfectly for loading data from REST APIs on Jira DC.
If you're looking for a Scriptrunner solution, you might want to look at Script Fields where you can script where to load data from for your field.
