Is it possible to to make a one-time change to rotation when someone is out sick?
You can use over-ride option by mentioning the particular time.

Yes, that is possible, and you do that by adding an override. You can choose to add another user as the override, or if you type in No one you basically remove that user from the schedule during the time you have selected. You can learn more about the override in this KB.
