How can I change the assignee field (avatar) of a column on an Agile board to a custom user picker field? I have gone through the board settings but couldn't find any option to do so. Is it possible to change it? Please see the attached screenshot for reference.

No, that is not possible.
You could add that other user picker field to the card layout to display it in addition to the Assignee.
