We are investigating the possibility of using Jira Service Management in the Data Center version.
We have a question regarding creating a user.
Is it possible to add profile fields so that, for example, when you create a new employee, you can directly store the supervisor?

Using native functionalities, storing additional information about users is impossible. As you're asking about JSM, I understand that "users" means "customers", isn't it?
To achieve it, you can use a paid app developed by my team called Customers & Organizations Management for Jira Service Management. With this app, you can define what properties you want to collect and assign values to every customer in the service project. For example, the supervisor, mobile phone, department, etc.
