I have a scenario where I create children tasks for a parent issue and I have created a link type of "assignment" with one direction being "is assignment of". I would like to restrict these tasks so that they can only have a single linke of "is assignment of". In this situation it doesn't make sense that the task could be an assignment to multiple parent issues. How can I enforce this?

You can do this only with some customization. You will have to write your own link operation and disable the system operation.
Once you write your own operation, in an add-on, you can add all kind of validations in it.
