Is it possible to access through VPN self-hosted JIRA Server with Integrated Mattermost?

Are external users enable to send messages and files in this "internal" Mattermost trough VPN?

Yes, Jira (and Mattermost) don't care if an incoming connection is via a VPN or direct, so you should be able to do this.
