I would like to make the target start and target end date required on Story's. Is this option possible?

Yes, of course. You can do so in several ways, however, depending on the type of project you are in and how this fits into your process.
Since you indicate being on a premium plan and are asking about target start and end dates, I assume you are using company managed projects. In that case, such fields are most commonly required at a certain point in the workflow. If you use company managed projects, it is best to add a fields required validator to the transition of your workflow where you want to make sure they are filled out. Make sure to add a transition screen to the same transition, so users can fill out the fields while they are changing the issue status.
In a team managed project, you can check the required box when you add a field to an issue type. The downside of that approach is that it is always required, even when you create an issue. On top of that, workflow rules can help you set restrictions on a field to, but afaik there is no option to check if a field has a value.

@Walter Buggenhout : Just to elaborate a little bit more on this. By using the Field Required Validator (in a company managed project) as I understand it only is triggered on the specific transaction on which it is added. Therefor when later (after that the transition has passed) maybe a user removes the value of that field this then will not be triggered upon unless in all the steps of the workflow a similar field required validation is added. Am I right?
Is there a way to make (Target) Start date and Target End Date (being locked fields) mandatory despite of the transition but applying to the issue type?
Hopefully I'm not missing anything in your previous answer to Barb, If I am please enlighten me.

You can use the validator on your workflow to make the fields required.
Go the project settings>>click on Workflow>>open the workflow which is assigned to Story issue type>> Edit the workflow>> Select a particular Transition>>click on validators>>add the validator by using the "Fields Required" option.>>then add the fields.

@Manoj Gangwar Is there any way to make these locked fields mandatory while creating the issue? As i would need a reporter to specify on the very start about Target start and Target end.
