Is it possible to do this?
For example, an employee needs to take a break or do other work
The employee presses the "I'm busy right now" button in Jira
And if someone tries to assign a ticket to this employee, the ticket is not assigned and a popup notification appears with the text that the employee added when he/she pressed the "Busy right now" button

Out of the box this feature is not available. This would be a good feature to have especially for Jira Service Management. There may be an app in the marketplace for this.
