We have a user that has been in a project for while with assignments etc. This user's roles have not changed. No new roles have been added. They suddenly don't show up in the list to assign an item to them!!! Even though they already have existing assignments. Why?
Could that account have been disabled by your admins? That won't affect their existing data, or even their groups or roles, it just makes them unselectable.
The account is active as far as I can tell. It has not been disabled. The user just updated a ticket that was assigned to them prior to this issue.

Ok, next thing to look at is the permission scheme - who exactly "can be assigned issues"?
