Example,

1) Let the field be "org". 

2) It should be mandatory when status moved from To Do to Done.

3)But it should not be mandatory while creating the issue or moving to other status

Yes, you can do this with the transition screen set between the transition of the two status.

And with the help of JSU Automation for Jira Plugin, you can put a validator to make fields required. Without Plugin its not possible.
