Suddenly our email notification functionality is not working. I have to manually flush out all emails.
Could you please let me know what could be the reason for this?
It was working without any issue till last week
This can be caused by multiple things and without checking the log files it's hard to ensure the correct root cause. I recommend you to raise a ticket at Atlassian Support and provide your JIRA logs.

If you wish, you can also check your logs yourself. Please take a look at the following file: JIRA_HOME/log/atlassian-jira-outgoing-mail.log
