You can add any number of user directories. So, the short answer is yes.

This doesn't the answer the question about how to connect to two domains and use both AD's in combination as a User Directory.

I can connect to one Domain and leverage it's LDAP, but now I need to connect to a different domain to get it's AD and leverage that.

You will need to add a separate user directory. JIRA will combine multiple user directories and you can provide permissions to users/groups from both.

If you have same usernames in both directories, the one from the first directory will be used.
